چکیده

Although lysergic acid diethylamide (LSD), the hallucinogenic agent, has been the subject of numerous investigations, little is known about its biologic fate. In recent studies Boyd et al.’ and St011 el ~1.2 have examined the tissue distribution of CL4-labeled LSD in mice. Stoll and his co-workers2 have shown that the Cr4-labeled material measured in tissues included LSD, as well as transformation products of the drug. Lanz, Cerletti, and Roth1in3 have also measured the tissue levels of LSD in mice by the use of a bioassay procedure based on the antagonism of LSD to serotonin-induced contraction of the uterus of the rat. These investigators, however, did not show that possible biologically active transformation products of LSD were excluded from the assay procedure. The development of a specific and sensitive method for the estimation of LSD in biological materials has enabled us to study the tissue distribution, excretion, rate of biotransformation, and metabolism of the drug, as well as the subcellular processes involved in its transformation.

Pharmacokinetics and Concentration-Effect Relationship of Oral LSD in Humans

BACKGROUND The pharmacokinetics of oral lysergic acid diethylamide are unknown despite its common recreational use and renewed interest in its use in psychiatric research and practice. METHODS We characterized the pharmacokinetic profile, pharmacokinetic-pharmacodynamic relationship, and urine recovery of lysergic acid diethylamide and its main metabolite after administration of a single oral...

Antagonism of histamine-activated adenylate cyclase in brain by D-lysergic acid diethylamide.

D-Lysergic acid diethylamide and D-2-bromolysergic acid diethylamide are competitive antagonists of the histamine activation of adenylate cyclase [ATP pyrophosphate-lyase (cyclizing); E.C. 4.6.1.1] in broken cell preparations of the hippocampus and cortex of guinea pig brain. The adenylate cyclase is linked to the histamine H2-receptor. Both D-lysergic acid diethylamide and D-2-bromolysergic ac...

Mechanism of lysergic acid diethylamide interference with rabbit antibody biosynthesis.

Lymphoid cells from hyperimmune rabbits producing antibodies to a hapten, incubated in the presence of d-lysergic acid diethylamide, continued to synthesize protein at a normal rate. Isoelectric focusing analysis of the low-molecular-weight protein secreted by the cells incubated with lysergic acid diethylamide indicated two components, with pI's of 4.9 and 5.2. Immune cells not exposed to lyse...
